USSING SYSTEM
An Ussing chamber is used to study solute transport across a confluent layer of cells. This layer is typically a monolayer of epithelial cells which form a barrier between the external and internal environments of the body such as those that line the airways, intestinal tract and bladder, but could also be a multilayered structure such as skin or 3D cell culture or a confluent layer of endothelial cells such as blood brain barrier in culture or corneal endothelium. Solute transport may be determined by measuring the flux of the solute or sometimes, in the case of charged solutes such as ions, by measuring the current across the tissue. These electrical parameters, together with tissue resistance, serve to assess ion transport and tissue integrity.
In order to accomplish these measurements Ussing systems have been designed to have three main components; namely, Ussing chamber, voltage/current clamp and data acquisition system.
1) The Ussing chamber is simply a device that enables one to place the tissue between two solution-filled compartments. The chamber may be fitted with electrodes for measuring voltage and passing current across the tissue.
2) The voltage current clamp is used to control either the voltage or the current across the epithelium and thereby determine bioelectrics such as transepithelial voltage, short-circuit current and resistance or conductance.
3) Results of the electrical measurements may be recorded using something as simple as a chart recorder or newer electronic versions of a chart recorder. Alternatively, it can be collected by an intelligent data acquisition system that acts as a chart recorder but also measures and plots tissue resistance or conductance in real time such as Acquire & Analyze.


EASYMOUNT USSING SYSTEM FOR CELL & TISSUE
The EasyMount Ussing Chamber System is based on the P2300 chamber, using the P23xx series slider to mount both cell culture inserts and a wide variety of tissues. Working chamber volumes are 2-7 mL (typical working volume is 5ml).

A voltage is generated across the epithelium as a consequence of ion selective channels in the membranes and active transport. This voltage is sensed by the black Ag/AgCl electrodes and measured by the voltage clamp instrument. Current may be passed by the clamp instrument across the epithelium via the white electrodes. When this current is sufficient to exactly cancel the voltage generated by the epithelium, the epithelium is “short-circuited” and the current necessary to do this is the short-circuit current (Isc).

When the solutions in the chamber compartments are the same, the Isc is a measure of the net active transport of ions across the epithelium. The P2300 Standard EasyMount Ussing Chamber: designed for studies using a variety of animal tissues and/or cell cultures. The chamber uses custom designed EasyMount Ussing Chamber Sliders (See the complete list of P2300 Series sliders) for mounting tissues or permeable membrane supports (Snapwell, Transwell, Millicell, Nunc). Usable half chamber volume is 2.0 – 8.0 ml. P2300 chambers are sold in pairs of two.

P2060 Dual Air Control Valve: The Dual Air Control Valve (Model: P2060) easily controls the amount of gas bubbling your chambers with fingertip control of the screw type precision air valves. Includes two needle valves in an aluminum housing. Mounts on all EasyMount chamber stands.

8-Channel Chassis / Multi-Channel Voltage Current Clamp VCC MC8
The Multi-Channel Voltage Current Clamp with OptiGain is a multi-channel voltage/current clamp designed to be a flexible tool for studying ion transport mechanisms by in vitro epithelial tissues.It provides the capability to control either the voltage or the current across epithelial tissues, thereby enabling one to measure electrophysiological responses to drugs, hormones, and/or ion substitutions as well as to compare measurements of ionic flux with the short-circuit current. The voltage/current clamp is suitable for use with both "leaky" and "tight" epithelia, tissue samples or cell culture, and works for a very wide range of tissue area.Each multi-channel voltage/current clamp has the included optiGain feature, which enables the user to optimize the instrument for the characteristics of the tissue under study by setting the current gain and fluid resistance compensation range from a pair of switches located on the front panel. This feature is unique to equipment by Physiologic Instruments.

The Multi-Channel Voltage Current Clamp offers these features:

Acquire & Analyze II data acquisition package for VCC MC8;
It consists of
-data acquisition hardware
- interface cables
-the ACQUIRE and ANALYZE software package.
Acquire & Analyze is designed specifically for transepithelial electrophysiology. It is not intended to be a general purpose instrument. While it records voltage and current from up to 8 voltage/current clamp channels, it also imposes a command signal across the epithelium and calculates and plots transepithelial resistance in real time. This is a significant benefit over other software packages which require post processing with manual input to calculate TEER at each time of interest. Data acquisition is easy to initiate and generally requires only selection of a new file name, entry of the surface area based on slider used, and a reference. Data collection rate can be quickly selected from 3 user defined (typically once every 1, 10, or 20 sec) speeds by clicking an icon on the main screen. This yields very small data files relative to those of other software packages where that system must continuously sample at a high enough rate to capture any pulse imposed by the clamp instrument and independent pulse generator.

Dummy membrane DM_MC6 Module: is an input module for connecting the voltage and current electrodes to the VCC MC2, VCC MC6, VCC MC8 and all expandable instruments. The DM MC6 is a small, shielded unit that should be mounted close to individual Ussing chambers and is recommended for use with both our EasyMount Ussing Chamber Systems or for other classical Ussing chamber systems. Each DM MC6 also contains a single channel “dummy membrane” which is useful for testing the the functionality of voltage/current clamp.

The DM MC6 may includes (1) AAA battery that will require replacing annually.
Connection from the voltage/current clamp to the DM MC6 is by a 4’ shielded cable with modular plugs part numbers 46-1000-4.One DM MC6 input module is required for each clamp channel.
