Axoclamp 900A is a powerful and easy-to-use instrument from Molecular Devices. It comes from the Axon Instruments® series of high‐quality, low‐noise microelectrode amplifiers. Like the Axoclamp‐2B that preceded it, the Axoclamp 900A offers several modes of operation that measure signals from single cells, tissue slices, and whole animal preparations. The advanced signal conditioning added to the Axoclamp 900A amplifier saves the expense of buying additional hardware and frees up valuable space in your electrophysiology setup.

By making the amplifier computer-controlled, several powerful new features have been added that make it easier to set up and run experiments and that keep experiments running longer. This exciting new instrument is designed to meet researchers’ needs today, as well as offer flexibility for future experiments.

Several modes of operation
This versatile amplifier offers several modes of operation, making it suitable for a wide range of biological preparations and for maximum flexibility in experimental design:

  • - Current‐clamp (I‐Clamp) mode, for measuring voltage responses, is available in two channels with independent Bridge Balance and I=0 options.
  • - Two‐electrode voltage clamp (TEVC) mode uses two microelectrodes, one for continuous recording of electrode voltage and the other for injection of current. TEVC mode can be used with large frog oocytes or smaller mammalian cells. The high output compliance of this mode makes it possible to voltage clamp currents that are larger than currents clamped with single‐electrode techniques.
  • - Discontinuous current clamp (DCC) mode can be used for more accurate voltage measurements, and is especially useful when small changes in electrode resistance occur during an experiment.
  • - Discontinuous single‐electrode voltage clamp (dSEVC) mode is used to voltage clamp small cells that cannot tolerate being impaled by two electrodes. Set up correctly, this mode eliminates the large series resistance inherent in many preparations.
  • - High‐voltage current clamp (HVIC) mode is used primarily for extracellular is applications.
 

Software Control
Instead of the usual front panel knobs and switches, the Axoclamp 900A is controlled by the Axoclamp 900A Commander, a program that runs on a Windows PC computer and communicates with the amplifier via a USB 2.0 interface. This control interface reports resistance, voltage, and current measurements and provides automation of Bridge Balance, Pipette Offset, and Pipette Capacitance neutralization. In addition it has “smart” features that protect cells from potentially damaging signal oscillations and automate mode changes based on internal signal thresholds or externally-applied signals. Amplifier settings such as gain, filter frequency, recording mode and input/output scale factors are automatically telegraphed to the pCLAMP 10 data acquisition software via the USB connection.

Excellent amplifier performance
The ±180 V output compliance used for TEVC and HVIC modes makes it possible to pass larger currents and ensures faster clamp speeds. TEVC and dSEVC modes both have wide AC voltageclamp gain ranges for excellent voltage control. When DC Restore is enabled, the DC voltageclamp gain is greater than 1,000,000, ensuring optimal voltage control for constant-voltage measurements. The new dSEVC design is more stable and twice as fast as the Axoclamp 2B Amplifier, providing an excellent alternative to standard continuous single-electrode voltage clamping
